ALPCO's Multimeric Adiponectin ELISA kit has excelled under the strenuous demands of the research community since 2005. Flexibility

ALPCO's Multimeric Adiponectin ELISA is the only ELISA available for measuring all the oligomeric forms of adiponectin in a single assay system. This classic "sandwich" ELISA employs two well-characterized monoclonal antibodies. These antibodies are precise and selective for monomeric and dimeric forms of adiponectin:

Human serum and plasma were treated by sample pretreatment buffer (1,2,3), and were separated by SDS-PAGE and analyzed with western blotting using capture MoAb-64405(A), detector MoAb-64404(B).

  • (1) 50mM Tris-HCl buffer (pH6.8) containing 2%SDS without boiling
  • (2) 50mM Tris-HCl buffer (pH6.8) containing 2%SDS with boiling
  • (3) Sample pre-treatment buffer in our kit without boiling

The antibodies detect only monomeric and dimeric adiponectin structures, which is not uncommon for an adiponectin ELISA. What separates the ALPCO Multimeric Adiponectin kit from other kits available, even others for quantifying high-molecular-weight (HMW) adiponectin, is the innovative sample pre-treatment procedure.

The two-step sample pre-treatment procedure first specifically digests adiponectin complexes that are NOT of interest in your study. For example, if you need to only quantify HMW adiponectin, the first step in the procedure digests mid- and low-molecular-weight adiponectin (Trimeric & Hexameric adiponectin, respectively), removing these oligomers from the sample, in 20 minutes. The second step reduces the remaining adiponectin oligomeric complexes into monomers and dimers, exposing the epitope sites for accurate measurement in the ELISA. To measure total adiponectin, just skip the digestion step, leaving all the protein intact until it is reduced from oligomers to monomers and dimers. A key benefit of this pre-treatment protocol is the flexibility it offers, allowing researchers to quantify adiponectin in all its circulating forms: total adiponectin, of course, but also to differentiate between circulating levels of trimeric, hexameric, or dodecameric and above (HMW).

Standardization

Prior to the introduction of this ELISA, multimers of adiponectin were measured using time-consuming laboratory techniques such as size fractionation and later, semi-quantitative western blots. The ALPCO ELISA correlates well to the Western Blot method, which remains a recognized reference method for HMW adiponectin measurement (3), but the ELISA can be completed in less than 4 hours.
